Effects of Praziquantel Treatment on the Hepatic Tissues of Albino Rats Infected with Clonorchis sinensis

Abstract
A histopathological Study was undertaken in order to observe the effects of praziquantel, which is known as the drug of choice against clonorchiasis, on the hepatic tissues of albino rats infected with Clonorchis sinensis. Praziquantel, at the total dose of 300mg/kg in a day, was administered to the rats of 9 experimental groups 5 weeks after the infection with 50, metacercariae of C. sinensis per rat. These rats were sacrificed group by group, which was classified by various time intervals after the drug treatment ranging from 6 hours to 5 weeks, and their hepatic tissues were removed and examined by conventional light microscopy.
The rats of non-treated control group, which were infected with C. sinensis but did not receive praziquantel treatment, were also sacrificed and examined at the same time intervals with the treated groups in order to compare the pathological changes of the hepatic tissue.
The results obtained were as follows:
1. The sectioned worms were recovered in the lumen of intrahepatic bile ducts, and this, finding lasted until 5 days after praziquantel treatment. But the worms were not monitored at all later than 1 week after the treatment, and thereafter the hepatic tissue failed to show any worm section in the lumen of bile ducts.
The earliest disappearance of the worm from bile duct occurred at 6 hours after the treatment.
2. The sectioned worms in the lumen of bile duct revealed extensive vacuolization in their parenchymal tissuse and epithelial destruction in their intestinal ceca, while their reproductive organs remained rather intact in spite of the drug treatment.
3. The rats in control group showed typical hepato-biliary changes of clonorchiasis, such as dilatation, thickening and adenomatous proliferation of biliary epithelia, fibrosis and. cellular infiltration of portal space and so on. But such pathological changes were gradually decreased in the praziquantel-treated rats especially after the disappearance of worms in tissue sections.
4. In the group of rats sacrificed 1 week after the treatment, hepatic tissue was almost restored its normal architecture except for slight -dilatation of the portal space. But this slight dilatation of portal space was also seen in the hepatic tissue of some rats even after 5 weeks from the treatment. On the other hand, complete recovery of the lesions occurred as early as 2 weeks after the treatment in some rats.
